THE ANALYSIS OF GENE EXPRESSION PROFILING FOR PHARMACODYNAMIC ACTION OF COMPOSITION OR COMBINATION OF JASMINOIDIN AND CHOLALIC ACID AFTER FOCAL CEREBRAL ISCHEMIA IN MICE

To elucidate the gene expression of focal cerebral ischemia mice responses to herbs treatment, we analyzed the expression of 372 mouse genes in focal cerebral ischemia before and after treatment with Jasminoidin and Cholalic acid given alone or in combination. We used generally accepted methods, including TTC staining, behavior test, as well as micro and ultra microscopy which can dynamically and accurately monitor pathological and physiological changes after cerebral ischemia on earlier period. Based on changes in gene expression, we identified 57 genes that accurately discriminated among the three treatments. Discriminating genes included those involved in apoptosis, signal transduction, cell cycle control and transporter activity. There are some differences in gene expression between Jasminoidin and Cholalic acid given alone and in combination, which is not simple accumulation of profiling of gene expression of single drug. These data indicate that gene expression of Jasminoidin and Cholalic acid after focal cerebral ischemia in mice share different pathways of genomic response to the same composition treatment, that changes in gene expression are treatment-specific and that gene expression can illuminate differences in biological response to drug combinations versus single agents.
