(tttta)n POLYMORPHISM IN THE PROMOTER OF GENE OF P450scc IN THE PATHOGENSIS OF POLYCYSTIC OVARY SYNDROME IN CHINESE WOMEN

To investigate the functional significance of microsatellite polymorphism (tttta) n in the promoter of gene of P450scc (CYP11a) in the pathogensis of polycystic ovary syndrome. Two hundred and one patients and 147 controls were studied. Menarche age was investigated. Body mass indices were determined. Blood samples were obtained for DNA analysis and hormone measurements. The genotype distribution of CYP11a is statistical difference between patients with PCOS and controls (Fishers Exact Test (2-Tail), p=0. 031). 4//8 and 6//6 genotype individuals were statistically different among patients and controls (1. 5% and 6.1%, respectively,X2= 5.467,p = 0.019; 57. 7% and 32. 8%, respectively, X2= 5. 588,p=0. 018). BMI of the 4//4 genotype patients is statistically lower than that of 4//6, 6//6, and 6//8 genotype patients (p=0. 003 7, 0. 002 7, 0. 059, respectively). No differences were observed in serum hormone concentrations among different genotype individuals in patients with PCOS. The allele (tttta)n distribution in the promoter of CYP11a is greatly different between Chinese Han woman and Spain, Greek, UK, or other Europid women and is statistical different between patients with PCOS and controls. Despite we did not find the difference with hyperandrogenism among the different genotypes of patients with PCOS, we hold that the (tttta) n pentanucleotide polymorphism plays a certain extent role in the pathogenesis of PCOS.
