A METHOD FOR DETECTION OF SEQUENCE SPECIFIC DNA-BINDING PROTEIN
Transcription factors played central role in drug development and disease diagonosis. There was a need for robust high throughput methods to analysis of transcription factors and sequence-specific DNA. Here unimolecular hairpin oligonucleotide probes were immobilized on polyacrylamide gel pad arrays and subsequently converted to double strand DNA arrays for detection of transcription factors. RhNF-kB p50 homodimer was detected by the miroarrays. The results indicated the method a economical, sensitive and efficient method and could be applied to analysis such as interaction between protein and sequence specific DNA as well as single nucteotide polymorphism site recognition.
