DEVELOPMENT OF COMPETITIVE IMMUNOASSAYS FOR MORPHINE DETECTION ON A PROTEIN CHIP

Competitive immunoassays on a protein chip as a detective tool for morphine were developed. An antibody chip based on the competition-restrain reaction principle between immobilized antimorphine and fluorescein-labeled BSAmorphine complex was developed with confocal scanner analysis method to detect the interaction of antibody and antigen of morphine. The optimal concentrations of antimorphine and BSA-marphine-Cy3 are 0. 031 3 mg/ml and 0. 012 5 mg/ml, respectively; the linearity range is 0. 01- 10 ng/ml. The limit of detection is 0. 001 ng/ml. The cross-reactions of antimorphine with methyl Benzedrine, amphetamine were not detected. But a certain extent cross-reaction between antimorphine and codeine was detected. The recovery of morphine and its metabolized products in tested urine is between 91. 2% and 109. 2%, with a detected limit of 0. 02 ng/ml. The result suggested that the antibody chip is a sensitive, specific, easy-operating, time-saving, high-throughput and sample/reagent-minimized method, and is useful in forensic medicine detection, effect monitoring during the process of abstaining from drug and drug-screening.
