AMPLIFICATION OF DNA DETECTION SIGNAL USING GOLD NANOPARTICLES AS AMPLIFIER BY QUARTZ CRYSTAL MICROBALANCE
Gold nanoparticles were utilized for amplifying the frequency signal of DNA detection using quartz crystal microbalance (QCM), DNA Immobilization and hybridization were monitored by QCM frequency shifts. Streptavidin-nanogold conjugations were connected with target DNA to amplify the frequency signal. It suggests that the frequency shift increases with the decrease of nanogold diameter and the increase of streptavidin-nanogold concentration. The optimal ratio of streptavidin-nanogold stock solution: PBS was 7 μ1:50 μ1. The sensitivity of this enhancement detection is 100 fM The complementary and mismatched target DNA could be distinguished clearly.
