DNA SEQUENCING BASED ON ELONGATION REACTION WITH A SERIES OF DEGENERATE PRIMERS ON MICROARRAYS
We have developed a novel method for sequencing DNA based on primer extension using a DNA poiymerase on microarrays. The templates sequences were detected by extending a series of degenerate primers that annealed to the template which were immobilized on the glass slides. The results show the dye-labeled nucleotide can be incorporated accurately in a base-specific manner into the primer after primer extension. This approach potentially can be developed into a high-throughput DNA sequencing system.
