MICROARRAY-IN-A-TUBE PCR ASSAY FOR HIV-1 DRUG RESISTANT MUTATION DETECTION
A novel microarray assay for the HIV drug resistance mutation detection was reported. In this assay, the microarray was arranged on the inner surface of a specially designed* optically transparent, plastic Eppendorf cap. The full analysis/inspection processes, including multi-(RT)-PCR, hybridization, and fluorescence detection, were carried out inside the encapsulated Eppendorf tube. The Microarray-in-a-tube assay PCR assay was successfully applied for detecting a number of HIV - 1 drug resistance mutations of HIV - 1 reverse transcriptase gene including K65R, K103N. V106A, V108I, Y181C, Y184M and Y188C. The assay system is low cost and simple to use, and prevents the false results associated with contamination of the PCR products.
