THE OPTIMAL CONCENTRATION OF FLUORESCENCE PROBE FOR REAL-TIME PCR ON DAN MICROARRAY
There are many kinds of methods used for quantitative PCR experiment. However, these methods can simultaneity detect 7 kinds of different gene fragments at best. Considering Microarray as a high throughput method could be detected many more gene site simultaneous. The Real-time PCR on DNA microarray developed in our laboratory. In this paper, we adopt the Gene Frames as the container of the PCR reaction on the microscope slides. During the PCR reaction, the fluorescence intensity of special fluorescence probe and non-special fluorescence probe (Mark) which were immobilized on the glutaraldehyde derived glass slide were be real-time detected by the scanner, and the change of the intensity of the special probe showed that the method is available. Through the experiment the optimal of the concentration of the probes was 5 μM. This result will be very useful for the further study of the Real-time PCR on DNA microarray.
