IDENTIFY AND SCREEN THE ASSOCIATED GENES AND POTENTIAL DIAGNOSITC MARKERS OF RAT PRIMARY LIVER CANCER

In order to study the difference of gene expression in Rats primary liver cancer (T) and normal tissue of Hver(N), and to screen for associated gene and diagnostic protein markers in early stage of primary liver cancer ( PLC) by oligonucleotide microarrays and bioinformatics, rat primary liver cancer were induced by DEN. and Affymetrix Rat Genenome 230. 2. 0. GeneChip was used to detect the gene expression profile difference in T and N tissue. Bioinformatics was undertaken to analyze the test results and screen diagnostic markers. As compared with normal liver tissue, a total 447 genes (EST/Full length) in primary liver cancers were found to be different with more than two fold in expression levels. Of the 447 genes, 271 were up-reguiated (Log ratio≥2), and 176 were down-regulated (Log Ratio≤-2). According to the Gene Ontology (GO) classification and Excel overlapping analysis, these genes were classified into differential biological process (204) and molecular function (178), 12 potential diagnostics protein markers have been screened out. In conclusion, tumor associated gene and potential diagnostics markers in early stage of primary liver cancer was successfully identified and screened by a combination of boinformatics and Affymetrix microarry technology.
