ACTIVATION OF SONIC HEDGEHOG PATHWAY IN UPPER-GASTROINTESTINAL TRACT SQUAMOUS CELL CANCER
Sonic Hedgehog (Shh) proteins are expressed during vertebrate developmentin some tissues and at epitheHal-mesenchymai boundaries in several developing organs, including the lung, gut, hair follicle, and tooth. In recent years, Shh signalling has recently been demonstrated in small celt lung cancer (SCLC). as well as in upper gastrointestinal malignancies arising from pancreas, esophagus and stomach. However, it is still unclear the function of Shh signaling in human upper-gastrointestinal tract cancer, including factors regulating the signal transduction pathway, identification of downstream target genes, and so on. Here we found that target genes of Sonic hedgehog pathway are elevated in 9 out of 15 primary esophageal squamous cell cancer and 3 out of 4 primary gastric squamous cell cancer. Activation of the pathway is found in upper-gastrointestinal tract squamous cell cancers and associated with poor differentiation and higher stages of the tumor. While the sonic hedgehog transcript is localized in the tumor tissue, expression of Glil and PTCH1 transcripts as well as sonic hedgehog protein are found both in the tumor and in the stroma, suggesting multiple mechanisms of hedgehog signaling during upper-gastrointestinal tract squamous cell cancer development Our data indicate that activation of the hedgehog pathway is an important event during development of upper-gastrointestinal tract squamous cell cancer.
