ALTERATION OF ppGALNAcTs GENE EXPRESSION IN THE 1, 25-DIHYDROXYVITAMIN D3 INDUCED DIFFERENTIATION OF HUMAN ACUTE MONOCYTIC LEUKEMIA CELL LINE SHI-1

To elucidate the role of ppGalNAcTs in the differentiation of SHI-1, we use 1,25 -(OH)2D3 as a inducer. In the present study, we have demonstrated that 1,25 -(OH)2D3 induced acute monocytic leukemia cell line SHIIdifferentiation into normal mature monocytes as confirmed by morphologic studies and analyzing CD11b expression on the cell surface. The real-time PCR results show that ppGalNAc-T1 increased after the cells were exposed to 1. 25 -(OH)2D3. The expression of ppGalNAc - T2 and - T3 was unaffected while ppGalNAc - T4 decreased We suggest that ppGalNAc - T1 and - T4 gene expression may be the molecular indicator in the differentiation of acute monocytic leukemia.
