CLONING, EXPRESSION AND PRIMARY FUNCTIONAL STUDY OF THE HUMAN β3GALT7 GENE, A NOVEL MEMBER OF THE β1,3-GLYCOSYLTRANSFERASE FAMILY

The β3GALT7 gene (AY277592, EC2. 4.1-) is a novel member of human β3-galactosyltransferase family isolated from human lung cDNA library In by our laboratory. To make further study, we expressed β3GALT7 protein in prokaryotic and eukaryotic system. Prepared anti-βGALT7 polyclonai antibody by immunizing rabbit with purified β3GALT7 protein. The expression of β3GALT7 in several tissues and cell lines was analyzed by Western blot and immunochemistry analysis. Subcellular location was observed using fluoroimmunocytochemistry assay. And we also made a primary study of its function related to tumors. As a result, we obtained the prokaryotic and eukaryotic recombinants and anti-β3GALT7 polyclonai antibody. Found β3GALT7 was located in cytoplasm around karyotheca. Immunochemistry analysis showed the enzyme was highly expressed in some tumor tissues and indicated an expression discrepancy between tumor tissues and normal tissues. The expression of MMP2 and MMP14 in transfected cells increased or decreased along with the expression of β3GALT7 suggested β3GALT7 may be also related to the soakage, metastasis and multiplication of tumors.
