EPITOPE EXAMINATION FOR ANTIBODY PREPARATION AND EXPRESSION ANALYSIS OF HUMAN POLYPEPTIDE: N-ACETYGALACTOSAMINYLTRANSFERASE-2 PROTEIN
O-glycosylation of proteins is initiated by a family of UDP-N-acetylgalactosamineipolypeptide N-acetylgalactosaminyltransferases (Ga1NAc-T). To study subcellular localization of ppGa1NAcT2, the secondary structure and surface properties of ppGa1NAcT2 was studied and we found that the prevailing epitopes of ppGa1NAcT2 existed in the RICIN domain, and we developed polyclonal antibodies against ricin of ppGa1NAcT2. These antibodies titer were 1:16000 detected by ELISA and they had good reactivity and high speciality in Western blot. We have localized ppGa1NAcT2 to Golgi apparatus by immunofluorescence. The results suggested that ppGa1NAc - T2 might take part in modifying glycoproteins in the Golgi complex.
