IDENTIFICATION OF GENES RELATED TO IN HUMAN PERIPHERAL BLOOD MONONUCLEAR CELL (PBMC) OF PATIENTS WITH HCV BY mRNA DIFFERENTIAL DISPLAY METHOD

In this study, the differential expression genes of PBMC of patients with HCV were firstly investigated by employing differential-display reverse transcription polymerase chain reaction (DDRT-PCR), Through compared gene expression profile of PBMC between 10 pairs of patients infected HCV and non-infected HCV controls, twenty-six differential fragments have been found and ten were confirmed by northern dot blot. We further have cloned, sequenced and done using the BLAST for homology matches in the GenBank database. Sequence results mainly indicate that those fragments are homologous with cytochrome c oxidaseI(MTCOI1), Mus musculus triggering receptor, Sus scrofa ubiquitin carboxyl-terminal hydrolase LI (UCHL1) mRNA, Homo sapiens phosphoinositide 3-kinase p85 subunit mRNA, Homo sapiens transient receptor, potential cation channel Sytl4 gene for synaptotagmin XIV. Among them, we are very interested in the phosphoinositide 3-kinase p85 subunit (PI3K), because the large data show that PI3K is closely associated with the NS5A of HCV and needs to further study.
