Effects of silencing TPX2 by siRNA on radiotherapeutic sensitivity of Hela cell in Human cervical Cancer
AIM: To investigate the effect of targeting protein for TPX2-specific siRNA on the raidiosensitivity of Hela cell in Human cervical Cancer.METHODS: The siRNA was introduced by Lipofectamine2000 into Human cervical cancer.RT-PCR and western blot was used to detect the expression of TPX2 mRNA and protein in Hela cells.Hela cells transfected were divided into 4 groups : control grouptransfected groupradiation group and combined treatmemt group.Four groups of cell were exposed to different doses of radiation (0,2,4,6,8Gy).Cell survival ratio was detected by means of CCK8 method.And the detection of the apoptosis rate and cell cycle was completed by FCM.
