The Identify and Production of Transgenic Mice and Goat with Capra hircus SCD1 gene
The crucial role of Stearoyl-CoA desaturase 1(SCD1)during goat(Capra hircus)fatty acid metabolism is corresponding with the composition of goat milk and muscle fatty acid.This study explored the expression of SCD1 gene in goat and detected the inherit and express of transgenic mice and goat,which demonstrate the possibility of production transgenic goat by Sperm-mediated gene transfer(SMGT).Method:The expression of SCD1 gene and fatty acid metabolic genes in goat mammary gland tissues and muscle tissues were obtained by GEO database,and detected by qRT-PCR.F1 transgenic mice were mated and then detected the target gene and protein expression level of F2 transgenic mice,simultaneously detected the content of the fat in F1 and F2 transgenic mice muscle.Moreover,production and identify the transgenic goat by testicular injection.Result:The results showed that the developmental-specific expression of SCD1 in the goat mammary gland tissue and muscle tissue.The positive rate of exogenic gene expression was 30%and exogenic protein expression was 6.67%in F2 transgenic mice,Moreover,the fat content of muscle in the positive group was significantly higher than the control group both in F1 and F2(p>0.05).At the positive rate of exogenic gene expression was 10.29%and exogenic protein expression was 8.82%in transgenic goat by testicular injection.Conclusion:This study investigated the role of SCD1 gene in fatty acid metabolism of goat and demonstrated the transgenic goat SCD1 mice could regulate the fat synthesis and steadily hereditary.The transgenic goat was successfully produced by testicular injection,which provide a reference for the production of transgenic livestock in breeding applications by testicular injection.
