基于多靶基因序列分析建立药品及环境污染葡萄球菌鉴定及分型溯源方法
Increasing clinical significance of coagulase-negative staphylococci (Co NS) requires to have more effective methods forspecies identification and genotyping of Staphylococcus.In this study, six new housekeeping genes (fem A, fts Z,gap,pyr H, rpo B, andtuf A) with extensive allelic polymorphisms were identified for developing a comprehensive MLSTscheme.The selected primers successfully amplified six loci from all of the 172Staphylococcus strains consisting of 18species. Sequence analysis of each locus (41 to 56 alleles) showed higher sequence diversity than 16S r RNA (26 alleles).Phylogenetic analysis of the concatenated sequence (3054 bp) of the loci provided species-level identification andhighly discriminatory typing for all the strains.Analysis of multilocus allelic profile of 172 strains generated 95sequence types(ST), suggesting a high degree of genetic diversity in the genusStaphylococcus,e.g. 30S.aureus, 37S.epidermidis, and 13S.hominis strains were typed into 15 STs, 21 STs, and 9 STs, respectively. Compared thecomprehensive MLST with previously developed MLST scheme, similar discrimination was obtained forS. aureus andS. hominis strains (S. aureus 17 STs and S. hominis 11 STs). Whereas for S. epidermidis, the loci inarc C,gtr, ortpi Awere failed to be amplified from threestrains by MLST primers.Taken all together, the comprehensive MLSTdeveloped herein provides more reliable results for species identification and genotyping ofStaphylococcus strains.
