LET7B INHIBIT THE WNT/β-CATENIN PATHWAY IN LIVER CANCER CELLS VIA DOWN-REGULATED FRIZZLED4 AND β-CATENIN
Let7b miRNAs are involved in carcinogenesis and tumor progression through their roles in maintaining differentiation and normal development.However,there is little research 2 focusing on the relevance between Let7b and Wnt/β-catenin signaling pathway.To study this,human liver cancer cells HUH7 and MHCC97H were cultured,enhanced or inhibited the expression of let7b in two cell-lines,measured the Wnt signaling related protein β-catenin and frizzled(FZD)family receptor,observed the proportion of CD133+CSCs in liver cancer cell lines,surveyed the proliferation,invasion and migration of liver cancer cells.The research revealed that enhanced expression of Let7b decreased the expression of β-catenin and FZD4,inversely inhibited Let7b expression increased β-catenin and FZD4 expressions.Enhanced Let7b expression reduced the proportion of CSCs in liver cancer cell lines meanwhile Let7b inhibition increased the proportion of CSCs.Up regulated Let7b expression repressed the proliferation,invasion and migration of liver cancer cells.This study showed Let7b repress the proliferation,invasion and migration of liver cancer cells and reduce the proportion of CSCs in liver cancers by inhibit Wnt/β-catenin signaling pathway via down-regulated FZD4 and β-catenin.
