Effects of Hyperbaric Oxygen Preconditioning on Caspase-Dependent Apoptosis in Neurons after Spinal Cord Injury in Rats

  Objective: Hyperbaric oxygen preconditioning (HBO-PC) can reduce neuronal apoptosis after spinal cord injury (SCI), but the mechanism remains unclear.In this study, we sought to investigate the effects of HBO-PC on motor function and cysteine-aspartic acid-specific protease (caspase)-dependent apoptotic pathways after SCI in rats.Methods: Thirty-six healthy Wistar rats were randomly divided intocontrol (n=12), SCI(n=12) and HBO-PC (n=12) groups.After 10 roundsof HB0-PC, the SCI modelwas inducedin both the HB0-PC and SCI groups.Two weeks later, motor function was determinedusingBasso Beattie Bresnahan (BBB) scoring, spinal MRI testing, spinal cord tissue section analysis and MEP testing.The spinal cord fromT7-9 was also collected to quantify apoptosis-related geneexpression.Results: The spinalcavity and atrophy levels, BBB scores, and MEP resultswere lower in the HB0-PC group than in the SCI group.Theexpression ofapoptosis-related geneswassignificantly increased in the SCI group compared with the CON and HBO-PC groups.Conclusion: HBO-PC can prevent the loss of motor function and nerve conduction due to SCI by modulating the endoplasmic reticulum and mitochondrial apoptosis pathways in neurons after HB0-PC.
