Comparative study on qualitative and quantitative analysis of Boschniakia rossica and its medicinal liquor using UPLC-MS/MS

  Objective: To optinize the extracting condition of Boschniakia rossica, control quality of medicinal liquor and compare the differences between raw material under optimum extracting condition and medicinal liquor of Boschniakia rossica.Methods: A Box-Behnken design along with a simple and quick method, high-performance liquid chromatography coupled with photodiode array detection was applied in order to obtain most content of phenylpropanoid glycosides and iridoids in Boschniakia rossica.Qualitative analysis of main components from medicinal liquor and raw material of Boschniakia rossica was studied through UPLC-MS/MS.Results: The optimum extracting condition was: time is 9 min, ratio of liquor to material is 108:1 mL/g, concentration of ethanol is 78%.And under this condition, the predicted yields of baschnaloside, rossicaside B and extraction were 23.7 mg/g, 2.8 mg/g and 31.3 %, respectively;five compounds were quantified in the liquor, the highest amount of them was rossicaside A, the second one was rossicaside B, then were baschnaloside, trans-p-coumaryl alcohol 1-O-β-glucopyranosyl (1→4)-a-rhamnopyranosyl (1→3)-β-glucopyranoside and triandrin;the data of UPLC-MS/ MS indicated significant differences between the medicinal liquor and raw material of Boschniakia rossica, more effective constituents in raw material.
