Nanosuspensions based nanogel as delivery system for nitrofurazone for dermal application

  Objective: The present investigation aimed for the development and characterization of nanosuspensions-based hydrogel formulations of nitrofurazone(NTZ)for enhanced topical delivery.Methods: The nanosuspensions were prepared using wet media milling and optimization by Box–Behnken design,The optimized nanosuspensions were subsequently incorporated into Carbopol 940 gel and characterized for particle size distribution,drug content,spreadability,pH,scanning electron microscopy(SEM),X-ray diffraction(XRD), in vitro release,ex vivo skin permeation and retention and stability.Results: The particle size analysis and SEM studies revealed NTZ nanosuspensions was still stably kept their particle size after suitably gelled by carbopol preparation.The drug content,pH and spreadability of the GLT nanogel was found to be 98.63 ± 2.2%,6.27 ± 0.2 and 25.74(g·cm)/s,which were within acceptable limits.XRPD studies suggested slight crystalline change in drug.Dissolution rate obtained from NTZ nanogel were markedly higher than NTZ-carbopol-gel.In vitro permeation studies through rat skin indicated that the amount of NTZ permeated through skin of NTZ nanogel after 24 hours was higher than NTZ-carbopol-gel,and NTZ nanogel increased the accumulative amount of NTZ in epidermis 4.5 times than NTZ-carbopol-gel.Conclusion:.These results suggested that the developed nanogel had a great potential as a viable approach to enhance the dermal bioavailability of NTZ.
