Immobilization of 5”-adenylic deaminase onto towel cellulose modified by p-β-sulfate ethyl sulfone aniline for improvement of enzyme properties
5”-Adenylic deaminase(5”-AD)from Aspergillus melleus was immobilized on cotton towels modified by p-β-sulfate ethyl sulfone aniline(SESA)and its functional properties were determined and compared with those of the free enzyme.The highest enzyme activities of free and immobilized 5”-AD were obtained in 0.2 M acetate buffer at pH of 5.5 and a temperature of 55C.An increase in Km(from 0.44 to 2.01 g·L-1)and Vmax(from 10.75 to 20.68 g·min-1·g-1)were recorded after immobilization.The apparent activation energy(Ea)of the free and immobilized 5”-AD was 30.03 and 24.43 kJ·mol-1,respectively,implying that the catalytic efficiency of the immobilized enzyme was restricted by mass transfer rather than kinetic limit.P-aminobenzene sulfate ethyl(ABSE)cellulose-5”-AD exhibited better thermal and pH stability than the free enzyme.The operational stability of ABSE cellulose-5”-AD was also investigated,and 54.46%of the original activity could be obtained after seven recycles,additionally,no significantly reduced in the activity was observed in the further three recycles,indicating that the ABSE cellulose-5”-AD has good reusability,and potential for industrial application.
