Enhanced Immune Response of Foot-and-mouth disease Type O Polypeptide Vaccine Using Poloxamer Adjuvant in Rats

Poloxamer is a thermosensitive polymer that exhibits solution–gel transformation as the temperature increases.The current foot-and-mouth disease(FMD)polypeptide vaccine that uses an oil emulsion adjuvant provides variable protection but induces injection-position side effects.This study aims to investigate the use of poloxamer as a potential adjuvant for FMD type O polypeptide vaccine to enhance immune responses in vaccine-treated rats.The sustained release properties of poloxamer are also investigated with the aim of prolonging the antigen residence time.We further investigate the capacity of poloxamer to increase splenocyte proliferation in vitro.Poloxamer increases cellular immune responses by generating IFN-γ,IL-2,and IL-6 cytokines.In conclusion,the FMD polypeptide vaccine formulated with poloxamer thermosensitive gel induces marked immune responses in vaccine-treated rats with the T cell phenotype.
