Preliminary studies on the relevance of antibodies and bioassay to experimental infection with Toxoplasma gondii in ducks
Little is known about the relationships between the dynamic antibodies and successful bioassay with Toxoplasma gondii infection in the ducks.The objective of the present study was to search the suitable opportunity of ducks infected with T.gondii for direct bioassay in mice.In this experiment,42 ducks were were allocated randomly into groups A,B and C with 14 birds in each group.At 14 days of age,the three groups were infected intravenously with 105,106 and 108 of T.gondii tachyzoites,respectively.On days 7,14,21,28,42,56,70 postinoculation(p.i.),sera were taken in each group to monitor the anti-T.gondii antibodies by modified agglutination test(MAT)method,and 2 ducks samples were used to bioassay for the T.gondii in mice.The T.gondii-like pathogens were identified by PCR and nucleic acids sequencing.In group A,the parasite was successfully bioassayed from day 7,14 and 21 postinoculated ducks,with the MAT titers 1:2,1:5 and 1:80,respectively.However,ducks with MAT titers 1:80,1:40,1:20 and 1:4 on the following days 28,42,56 and 70 were failed.In group B,T.gondii was successfully biossayed at all the timepoints,regardless of the seropositive or seronegative for T.gondii.Group C animals were all died before day 4 p.i,and they were not used for bioassay.PCR and sequencing results confirmed that all the isolates belonged to T.gondii.This study demonstrates that successful bioassay is incompletely depending on the positive MAT diagnosis.The existence of T.gondii and the amount of the parasite may be the key parameters during the bioassay studies.
