Molecular cloning,characterization,and tissues expression analysis of the goose(Anser cygnoides)prepro-VIP gene
In avian,vasoactive intestinal peptide(VIP)as a prolactin(PRL)releasing factor,is involved in the control of the PRL release,and plays a pivotal role as a regulator of reproductive behavior and neuroendocrine secretion.In this study,a 833 bp fragment covering the complete coding region(CDS)of goose prepro-VIP gene was obtained.The cDNA contains a 32 bp 5′-untranslated region(UTR),a 306 bp CDS,and a 495 bp 3′-UTR including a TGA termination codon(nucleotides 518–520 bp)and the poly(A)tail.The goose prepro-VIP gene shares about 67–94%nucleic acid identity and 53–98%amino acid identity with their counterparts in other species.The sequence analysis indicated that two alternatively spliced transcripts were identified,the larger transcript(VIP-1)encoded both VIP and peptide histidine isoleucine(PHI)exons and the short one(VIP-2)only encoded VIP.Based on the cDNA sequence,the genomic structure of this gene was characterized.The goose prepro-VIP gene consists of 7 exons and 6 introns,just as that in other avian species.The RT-PCR analysis indicates that the level of the VIP-1 is much higher than that of the VIP-2,and the lager one negligible or absent in liver,kidney,muscle and abdominal fat,while the short one is widely distributed in all examined tissues.
