The research of anti-tumor mechanism of ophiopogonin D on MCF-7 cells

Microtubules are one of the major cytoskeletal components and play critical roles in diverse cellular processes, cell division and mitosis.It has become a successful target for anticancer therapy.Previous studies have shown that large amount of natural products or synthetic drags could interact with microtubules and interfere with the function of microtubules.Ophiopogonin D is one of the main chemical compositions of Ophiopogon japonicus which has variety of pharmacological activity.Here we measured the inhibitory effects of Ophiopogonin D on the growth of MCF-7 breast cancer cells.The experiments include MTT, Colony formation assay, cell cycle, westem-blot, Tunel, Tubulin Polymerization Assay, Immunofluorescence microscopy, Molecular docking.Our results showed that Ophiopogonin D could inhibit the growth of cells, arrest the cells in G2/M phase and induce the cell apoptosis.Further mechanism studies proved that Ophiopogonin D induced cells apoptosis by regulating protein expression of Caspase family and promoted the polymerization of tubulin.The findings of this study indicate that Ophiopogonin D may be used as a new microtubule-inhibitor.
