Improving CRISPR/Cas9 multiplex editing ability by using the endoribonuclease Drosha

  Higher organisms, such as mammalian animals and plants, usually have a complex network to organize all their life activities.Thus multiple targeting in genome is of great meaning for both scientific research and clinical use.The bacterial type Ⅱ clustered regularly interspaced short palindromic repeat (CRISPR) and CRISPR-associated (Cas) protein system has been engaged in introducing double-strand breaks (DSB) to a specific target.But it is still a challenge for multiple genome targeting in animals.Here we report a Drosha-based multiple-sgRNA expressing method.Using this method, we got the multiple mutations in each different targeted genome loci.Because of the universality of Drosha in eubacteria and eukaryotes, this method could make the CRISPR/Cas9 system much more efficient and applicable in genome editing.
