Effect of BMP-7 on the expression of mesenchymal-epithelial transition markers insilicosis model
This study presented the effect of bone morphogenic protein-7 (BMP-7) inhibiting epithelial-mesenchymal transition (EMT) in silicosis model In vivo, Wistar rats were exposed to silica by intratracheal instillation and 7 days later rats were treated with BMP-7 or saline every other day.Rats were sacrificed at 15 days and 30 days after instillation to observe the expression of Vimentin, E-cadherin, TGF-β and phosphorylation-p38 MAPK in the lung tissues.Immunohistochemistry demonstrated the increasing expression of Vimentin and decreasing expression of E-cadherin, however, after intervention of BMP-7 Vimentin expression was down-regulated and E-cadherin expression was up-regulated.Western blot analysis showed the expression of TGF-β and phosphorylation-p38 MAPK in BMP-7 group were lower than that in silica group.In vitro, we found that the occurrence of EMT in A549 cells stimulated by silica could be inhibited by BMP-7.Our data showed BMP-7 inhibited EMT induced by silica associated with inhibiting TGFβ/p38 MAPK pathway.
