The Anti-inflammatory and Anti-osteoarthritis Activities of Pyrola Calliantha Extracts

OBJECTIVE: This study aimed to investigate the anti-inflammatory and anti-osteoarthritis activities of Pyrola calliantha H.Andr.(PC) in vivo and in vitro.METHODS: Carrageenan-induced hind paw edema and xylene-induced ear edema models were the two acute inflammatory rat models investigated.Rat knee osteoarthritis (OA) model was also duplicated for synovial fluids (SF) preparation.The levels of pro-inflammatory cytokines, IL-1β and TNF-α, in SF were determined by ELISA.Histological examination was employed to determine the pathological changes, including apoptosis of chondrocytes.The apoptotic rate of separated cultured chondrocytes was also determined by fluorescent staining.Expression of MMP-13 and Bcl-2/Bax at mRNA and protein levels were evaluated by qRT-PCR and Western-blot,respectively.
