The quantitative real-time PCR analysis of main methanogens in pit mud of different pit age

Methanogens are important functional miacrobes in pit mud for the fermentation of Luzhou-flavor liquor.The aim of this work was to investigate the regularity of main Methanogens in pit mud of different pit age.This paper made quantitative analysis of Methanosaeta and Methanosarcina in pit mud with cellar age of 5, 30 and 50 years, respectively, by using real time quantitative PCR assay.The results show that the abundance of Methanosaeta and Methanosarcina increase in varying degrees with the pit age rising of pit mud.This research analyzed the copies of main Methanogens in pit mud of different pit age by using quantitative PCR assay for the first time, laid an initiai foundation for identifying good quality pit mud, and provided a theoretical basis for understanding the Mechanism of microbial fermentation of Luzhou-flavor liquor.
