A method to verify dominant bacteria in clone library by application of qPCR

　　The quantitative PCR (qPCR) method was used to verity the dominant bacteria in fermented grains” clone library.According to the clone library, the Lactobacillus acetotolerans was the dominant bacteria and its percentage of clones in total effective clones was 73.30% in the 10 weeks” fermented grains.The qPCR experiments were conducted to quantify the 16S rDNA copy number of L.acetotolerans in PCR purified product 1, meanwhile, the total 16S rDNA copy number of total bacteria from PCR purified product 1 was examined by biological spectrophotometer.The results showed that the total 16SrDNA copy number of total bacteria and L.acetotolerans were 7.47 × 1012 copies/mL and 5.64 × 1012 copies/mL, respectively,therefore, the percentage of L.acetotolerans was 75.50%.It means that application of qPCR can verity dominant bacteria in clone library.This paper offered a quick and simple method to confirm dominant bacteria in clone library.
