Hepatoprotective effects of tocotrienols on lipid peroxidation, antioxidant scavenging enzymes, and PPAR expression in the liver of high-fat diet-fed mice
Scope Prolonged consumption of high fat diet primarilyaffects the liver thru the production of ROS that causes oxidative stress.Tocotrienols are antioxidants belonging to the vitamin E group.However, evaluation of tocotrienó1s physiological effects is very few compared to tocopherols.This study evaluated the hepatoprotective effects of tocotrienols on the liver of high fat diet-fed mice.Specifically, its effect on lipid peroxidation, antioxidant scavenging enzymes and expression of PPAR α and γ were analysed.Methods and Results Kunming mice wereuniformly divided into 6 groups.The first group served as normal control while the second group served as high-fat group.Group 3 served as 2nd normal control given with tocotrienol (200mg/(kg · day)).Groups 4,5 ,and 6 served as treatment groups given with high fat diet and different doses of tocotrienols (200,100, and 50 mg/(kg · day), respectively).Tocotrienols greatly reduced lipid peroxidation while influences the total antioxidant capacity of the liver.It also increased antioxidant scavenging enzymes and increased the expression of PPAR α and γ.Conclusion Fat accumulation in the liver can initiate chains of events the negatively affects its integrity.However, tocotrienols, as a hepatoprotective compound, acts in more ways than one.It not only counteracts oxidants but also helps in the expression of other oxidant-neutralizing compounds and genes that help regulate the cells metabolism.
