Inhibition of NF-κB and CCR7/CCL19/CCL21 contribute to the attenuation of inflammation by Baicalin in OVA-induced asthmatic mice
Baicalin, extracted and purified from the Chinese medicinal plant ScutellariabaicalensisGeorgi (Chinese name: Huang Qin) exhibits potent anti-inflammatory activity against asthma.However, it remains unknown whether baicalin can inhibit CC-chemokine receptor 7and its ligands important for initiating airway inflammation.Herein, we investigated the effects of baicalinon CCR7 and its ligands CCL19 and CCL21 as well as NF-κB pathway in asthmatic mouse model.Acute asthmatic model was established by ovalbumin (OVA) intraperitoneal injection and inhalational challenge.Within24h of the last OVA challenge, lung function was detected by direct airway resistance analysis.Lung tissues were examined for pathological changes.Inflammatory cells counts in broncho alveolar lavage fluids (BALF) were assessed.ELISA was utilized to evaluate the OVA-IgE, CCL19 and CCL21 levelin BALF.IL-6 and TNF-αlevel in serum were also detected by ELISA assay.Protein expression of chemokine receptors CCR7 as well as phosphorylation-IκBα (p-IκBα)and phosphorylation-p65 (p-p65)were tested by western blot and qPCR was used to determine CCR7 mRNA.Our data showed that oral administration of baicalin significantly improved pulmonary function and attenuated inflammatory cells infiltration into the lung, the levels of OVA-IgE,IL-6 and TNF-α,CCR7 and its ligand CCL19 as well as NF-κB were prominently suppressed by baicalin.The CCR7 mRNA levelwas substantially reduced.Our results suggest that baicalin has an inhibitory effect on airway inflammation, and it might be associated with inhibition of CCR7 and CCL19/CCL21, which may provide new mechanistic insights into the anti-inflammatory effects of baicalin.
