Ginsenoside Rd therapy improves histological and functional recovery in a rat model of inflammatory bowel disease Running title: GRd improves histological and functional recovery

  Backgroud: Ginsenoside Rd(GRd) is a bioactive constituent of the herb ginseng that has been reported to stimulate endogenous stem cell proliferation.The purpose of this study was to explore the effect of GRd on GI mucosa regeneration in a rat model of inflammatory bowel diseases (IBD).Methods: A rat model of indomethacin-induced IBD was established and administered orally with GRd at 10-40 mg/kg daily for 7 days.The effects of GRd were evaluated by morphometry of intestinal mucosa, double labeling of 5-bromo-2-deoxyuridine (BrdU)-positive cells with endogenous proliferative and intestinal stem cell markers, and polymerase chain reaction (PCR) analysis of the molecular markers in the treated rats.Results: BrdU was found in the recipient mucosa, partially co-localizing with intestinal stem cell markers Bmi, Lgr5, and Musashi-1.The mRNA levels ofBmi, Lgr5, and Musashi-1 were significantly increased in the intestine by GRd.The mucosal and crypt layers of the small intestine were thicker in the GRd-treated rats.The most significant changes in the observed parameters were seen in the 20 mg/kg treatment group.Conclusions: We demonstrated that GRd stimulates the intestinal stem cell proliferation in a rat model of IBD, resulting in improved intestinal functional recovery.
