The establishment of cognitive impairment rat models induced by chronic poisoning of organophosphate
Objective: To explore the reasonable administration way and dose of omethoate in cognitive impairment rat models induced by chronic poisoning of organophosphate.Methods: All rats were daily administered omethoate by gavage way (G) and subcutaneous injection way (S) with low, medium and high doses.The general condition and survival situation of rats in each group were observed during the experimental period.The memory reappearance test was operated by Y-electric maze after the model established.The morphology and structure of hippocampus and liver were observed by HE staining.Results: During the experiment, rats in each exposure group showed mild poisoning symptoms.The poisoning symptoms of rats in group G3 and S3 were most obvious, followed by group G2 and S2 and mildest in group G1 and S1.In group G1, 7 rats died with survival rate of only 30% and 5 rats died with survival rate of 50%, so group G3 and S3 (10mg/kg) did not suitable for establishing models.The results of the memory reappearance test showed that the memory keeping ability (MKA) of rats in each exposure group declined at different degrees, and rats in group S2 (5mg/kg) for 4 weeks were more obviously.HE straining showed that liver and hippocampus in each exposure group was damaged at different degrees compared with normal saline group.The denatured cell index (DCI) showed that rats in group S1 (2.5mg/kg) had the mildest damage to liver, while in group S2 (5mg/kg) had the medium damage to hippocampus.Conclusion: According to the synthetical analysis for general situation, survival rate,MKA and DCI of liver and hippocampus, Group S2, subcutaneous injection with 5mg/kg each day for 4 weeks was one of the reasonable administration way and dose to establish cognitive impairment model by chronic poisoning of omethoate in rats.
