Conditional Knockouts Generated by Engineered Endonuclease Reveal Roles of Coronin in C.elegans Neural Development
Animals with conditional gene knockouts are valuable tools for studying mechanisms underlying cell and developmental biology.We have developed a conditional knockout strategy by spatiotemporally manipulating the expression of an RNA-guided DNA endonuclease,CRISPR-Cas9,in C.elegans somatic cell lineages.We showed that somatic CRISPR-Cas9 technology offers the highest possible speed to efficiently create conditional knockouts in various cell types at different developmental stages.
