Dynamic monitoring of the cytotoxicity effects of protoberberine alkaloids from Rhizoma Coptidis on HepG2 cells using the xCELLigence system

  Rhizoma Coptidis is a traditional Chinese medicine mainly consists of protoberberine alkaloids(PAs).Recent research revealed the various pharmacological activities of Rhizoma Coptidis,and the antitumor activity of the PAs received more and more attention.However,most studies have focused on the effects of berberine while the studies about the other PAs still remain poor.In this study,we conducted a rationalized screening to investigate the cytotoxicity effects of the six PAs from Rhizoma Coptidis on HepG2 cells in vitro using the impedance-based xCELLigence system,and evaluated the applicability of the system by comparing it with traditional methods.Our study provided a reliable and efficient tool for real-time screening of compounds with antitumor activity.The present data showed that all of the six alkaloids could inhibit cell growth and induce death in HepG2 cells in a time-and concentration-dependent manner,and coptisine had comparative cytotoxicity effect with berberine.Moreover,combined with previous studies,we further analyzed the effects of the PAs on HepG2 cells and their structure-activity relationship.
