Cloning of a Novel Chitinase Gene from Paecilomyces lilacinus
Cloning and characterizaion of a novel chitnase gene Plchi1 from Paecilomyces lilacinus is described.The full length cDNA of Plchi1 containing a 1284 bp open reading frame (ORF) that could be translated into a 427 aa amino acids polypeptide.A signal sequence cleavage site located between aa 25 and 26.Additionally,Plchi1 contained two highly conserved regions of the family 18 glycosyl hydrolases, i.e.the potential chitin-binding domain (IMLSIGGW) and the catalytic active site (FDGIDIDWE).Phylogenetic analysis showed that Plchi1belonged to the class V fungal chitinses and was similar to other endochitinase genes from mycopathogens,entomopathogens and nematopathogens.
