Study on antimicrobial resisitance and drug resistant genes in clinical isolates of Acinetobacter baumannii
Objective: To investigate the distribution and drug resistance of Acinetobacter baumannii for appropriate application of antimicrobials.To investigate the positive rate of biofilm related genes and IntI1 gene among clinical isolated strains of Acinetobacter baumannii and analyze the relationship between them and drug resistance.Methods: The drug resistance of clinical isolated strains of Acinetobacter baumannii from Jan.2008to Dec.2010 in Intensive Care Unit were analyzed retrospectively.72 strains of Acinetobacter baumannii in Qingdao Municipal Hospital from Sep 2011 to Jul 2012 were collected randomly.bioMerieux VITEK 2 automatic microbial analysis was applied to species identification and drug susceptibility test.PCR assay was applied to detecting biofilm formation related genes-3 genes of the pili assembly system, outer membrane protein gene ompA, Extended-spectrum beta-lactamases gene PER-1and Autoinducer Synthase gene abaI.We detected the positive rate of IntI1 gene at the same time.Results: The resistance of Acinetobacter baumannii from 2008 to 2010 in ICU showed the rising tendency to all kinds of antimicrobials, except to GEN and SXT.The isolation rate of PDR-AB were4.35% in 2008 and increased to 39.7% in 2010.Of all Acinetobacter baumannii strains, csuC, csuD,csuE, ompA, blaPER-1, abaI and IntI1 gene amplifications positive number (%) were 61 (84.7%),58 (80.6%), 51 (70.8%), 66 (91.7%), 8 (11.1%), 54 (75.0%), 45 (62.5%), respectively.AbaI and IntI1had significant correlation with drug resistance(P < 0.05).Conclusions: The antimicrobial resistance of Acinetobacter baumannii in Intensive Care Unit is serious during the period of 2008 to 2010.The isolation rate of PDR-AB was higer year by year.High incidence of biofilm formation related genes and IntI1 were found in our hospital Acinetobacter baumannii isolates.AbaI and IntI1 could be closely related to drug resislante of Acinetobacter baumanni.clinicians should pay attention to the biofilm resistance mechanism.
