Short-term safety and effects of a novel fully bioabsorable poly-L-lactic acid sirolimus-eluting stents in porcine coronary arteries
Background Fully absorbable stents may have positive effects on arterial healing, and reduce the need for prolonged antiplatelet therapy. This study aimed to test the safety and efficacy of a novel fully bioabsorbable PLLA sirolimus-eluting stents (SES) in porcine coronary arteries. Methods Twelve PLLA SES and twelve Cypher SES were randomly deployed in the coronary arteries of healthy minipigs.Animals were sacrificed at 28 days, and the vessels were fixed and processed for histochemistry. Results At 28 days, there was a reduction in neointimal proliferation by PLLA SES compared with Cypher SES(neointimal thickness =(0.14±0.08) mm for PLLA SES vs.(0.30±0.14) mm for Cypher SES, P=0.002; % area stenosis =(23.25±15.99)% for PLLA SES vs.(39.09±21.01)% for Cypher SES, P=0.053).There was no active arterial wall inflammation, hypersensitiveness, necrosis, or intraluminal thrombus in either group. Conclusions The current study demonstrates that the fully bioabsorbable PLLA sirolimus-eluting stent is safe and feasible in the prevention of neointimal hyperplasia at 28 days.Further studies are required to examine its long-term effectiveness and safety.
