Atorvastatin reduces apoptosis of human coronary artery endothelial cells induced by advanced glycation end products through PI3K/AKT/mTOR pathway
Background The aim of the study is to study the mechanism of atorvastatin reducing advanced glycation end products(AGE) induced apoptosis of human coronary artery endothelial cells (HCAEs).Methods There were four groups, (1) Blank control group; (2) AGE group: HCAEs were incubated with AGE (0.1g/L) for 24 hours; (3) AGE+atorvastatin group: HCAEs were incubated with concentration of atorvastatin (1 μmol/L) for1 hour, then incubated with AGE (0.1 g/L) for 24 hours; (4) PI3K inhibitor (LY294002) group: HCAEs were incubated with LY294002 (5000 nmol/L) for 1 hour, then incubated with atorvastatin (1 μmol/L) and AGE (0.1 g/L) for 24 hours.Then, HAECs was stained by ANNEXINV/PI and ANNEXINV/CD144 endothelial microparticles were analyzed by flow cytometry; Western blotting was performed to detect PI3K, p-AKT, p-mTOR protein.
