Long noncoding RNARP5-833A20.1inhibitsproliferation and metastasis and cell-cycleby suppressing NFIAexpression in U251 cell
Backgrounds& Aims:Early reports suggest that nuclear factor IA (NFIA) plays important roles in glioma pathogenesis.Our recent study demonstrated that Long noncoding RNA (1ncRNA) RP5-833A20.1 suppressed the expression of NFIA in THP-1 macrophage-derived foam cells.However, the effect and possible mechanism of RP5-833A20.1 on glioma remain unclear and whether NFIA-dependent pathway involving in this progress has not yet been explored.Here, we investigated the mechanisms by which the RP5-833A20.1 regulatesthe expression of NFIA in glioma.Methods:The expression of RP5-833A20.1 and NFIA was determined from U251 cells and clinical samples by RT-qPCR.The effects of RP5-833A20.1 on cell proliferation, invasion, cell cycle, and apoptosis were evaluated by in vitro assays.The potential protein expression changes were investigated by Western-blotting.The methylation status of the CpGisland in NFIA promoter was explored by bisulfite sequencing.Results:We found that RP5-833A20.1 expression was down-regulated, whereas NFIA expression was up-regulated in glioma tissues compared to corresponding adjacent nontumor tissues from 20 glioma patients.Overexpression of RP5-833A20.1 inhibited proliferation and cell-cycle progression, and induced apoptosis in U251 cells.Both mRNA and protein levels of NFIA were markedly inhibited by overexpression of RP5-833A20.1 in U251 cells.Overexpression of RP5-833A20.1 increased the expression of miR-382-5p in U251 cells.BSP assay revealed that over-expressing RP5-833A20.1 enhanced the methylation level of NFIA promoter.Concluions:This study demonstrates that RP5-833A20.1 inhibits tumor cell proliferation, induces apoptosis, and inhibits cell-cycle progression by suppressing NFIA expression in U251 cell.Collectively, these results representRP5-833A20.1as a novel therapeutic target for glioma.
