Association analysis of single polymorphism variants in SOD1, SOD2 and GPx-1 genes with sperm DNA damage in a Chinese population
Oxygen stress is one of the causes of sperm DNA damage. In order to find some useful markers to predict the sperm DNA damage, we evaluated 9 single nucleotide polymorphisms (SNPs) variants of the SOD1, SOD2 and GPx-1 genes in a Chinese population. We compared the minor allele frequency (MAF) of each SNPs between the controls and the cases with sperm DNA fragmentation index (DFI) more than 30%, and found that SNP rs2842980 of SOD2 gene presented a significant difference (P=0.041, OR=0.824, 95%CI: 0.604-1.124), however, this difference failed to reach statistical level after Bonferroni correction. For all the other SNPs, the genetic variants between the controls and the sperm DNA damage cases did not reach any statistical difference. We performed further haplotype analysis in 3 blocks of the 9 SNPs, none of the haplotypes indicated a statistical different distribution between the controls and the cases. In conclusion, the polymorphisms rs2842980 of SOD2 gene might be a potential genetic marker for the sperm DNA damage in the Chinese population, however, further evidence is needed to confirm its values.
