The novel Polymorphisms in 5” Regulation Region of GPR54Gene and their Association with Sexual Maturity in Cattle

  GPR54 has been implicated in the neuroendocrine regulation of GnRH secretion.To determine the polymorphism of GPR54 gene and its association with sexual maturity in cattle,Simmental with early sexual maturity and Chinese native Wanbei cattle with late sexual maturity were cross bred.Cultivate cattle by Simmentai and Wanbei parents were selected for analyzing DNA polymorphism in 5” regulation region of GPR54 gene and their sexual maturity.The detecting fragment(GPR973) located in region from-1bp to-973bp upstream from the CDS start of GPR54 gene.The results showed that GPR973 contained a potential GPR973-1 promoter and its upstream region(GPR973-2).Two polymorphic sites, C-816T and T-754C, were found in GPR973-2.The DNA polymorphism analysis uncovered that Simmental cattles were only with the 816CC754TT genotype and Wanbei only with the 816Tr754CC genotype.The genotypes in cultivated cattle group were the combination of Simmental and Wanbei cattle.The average months of first oestrus age of cows (FOA)among genotypes in cultivated cattle group significantly differed, and FOA of cattle with the816CC754TT genotype decreased 1.28 months than that with the 816TT754CC genotype (p<0.01).The results of dual-luciferase reporter assays indicated that the 816C→816T and the 754T→754C could weaken gene expression (p<0.01).These studies have showed that the polymorphism at C-816T and T-754C sites played important role in cattle sexual maturity, and can be employed for candidate marker in cattle breeding.
