Generation of Transgenic Luxi Cattle Expression of hFat-Ⅰ
The fatty-acid desaturase enzyme could convert n-6 polyunsaturated fatty acids into ω-3polyunsaturated fatty acids (ω-3PUFAs), which found mainly in fish Oils, recognized as essential in preventing many diseases, such as coronary artery disease, cancer, diabetes, arthritis and depression.The use of transgenic cattle over expressed fatty-acid desaturase will promote people”s healthy diet.Herein, we constructed a vector contained codon optimized C.elegans hFat-1 gene and transfected into fibroblasts.Transgenic embryos were reconstructed by nuclear transfer of positive cells into enucleated in vitro matured oocytes.A total of 94 recipients were transplanted in two lots, 20 individuals were pregnant and the pregnancy percentage was 21.3%.4 calves have been born, two of them had already been dead during delivery and the other two died within 15 min and 88 h respectively.There are still 15cattle pregnancy.According to the results of PCR and sequencing analysis, three of the four were demonstrated to be transgenic cattle.Besides, the results of RT-PCR indicated that the hFat-Ⅰ gene expressed in liver, muscle, kidney, lung, as well as in heart in the transgenic cattle.
