Association of Adiponectin gene (ADIPOQ) rs2241766 Polymorphism with Obesity: A Meta-Analysis
Background Adiponectin plays an important role in regulating glucose levels and fatty acid oxidation.Multiple researches had focused on the association between rs2241766 polymorphism in the adiponectin (ADIPOQ) gene and obesity susceptibility. However, the results are inconsistent and inconclusive. The aim of this meta-analysis was to investigate this association.Method Literatures were searched from Medline, Pubmed, Elsevier, Chinese Wan Fang database and China National Knowledge Infrastructure (CNKI) databases. In this meta-analysis, odds ratio (OR) and 95% confidence interval (CI) were calculated with random-effect model or fixed-effect model depending on the presence of the heterogeneity.Sensitivity analysis was conducted to confirm the reliability and stability of the meta-analysis.Egger”s test was adapted to assess publication bias. Results In total, 13 relevant studies with 1875 obese and 1801 controls were included. The rs2241766 polymorphism was associated with obesity susceptibility in Chinese population, and T allele was considered to be the risk allele.Significant associations were found in the four genetic models (in allelic model: OR=1.28, 95%CI: 1.14-1.43;in additive model: OR=1.63, 95%CI: 1.25-2.14;in dominant model: OR=1.33, 95%CI: 1.01-1.75, and in recessive model: OR=l.51, 95%CI: 1.17-1.95, respectively). While in non-Asian population, we also found a weak association in allelic model, that is subject with the G allele had 0.73-fold (95%CI: 0.54-0.98) decreased risk for obesity. There was no significant evidence of publication bias. Conclusion Results of this meta-analysis demonstrated that rs2241766 polymorphism was associated with obesity. The results in different ethnic population may be different.More well designed studies with large sample size focusing on ADIPOQ -rs2241766 polymorphisms and obesity are required to confirm our findings in the future.
