Mutation by Co60 Irradiation Method and Fermentation of Trichoderma Viride for High Cellulase Activity
Trichoderma viride 3.2941 was mutated by Co60 irradiation method and selected by methyl-red plate with cellulose.13 mutation strains which had lager hydrolysis cycles were semi solid state fermentated,then selected using filter paper activity (FPA) as an index.Cellulase production conditions of one of the mutant were optimized by single factor and orthogonal tests in solid state fermentation.The results indicated that at conditions of 3g corn cob, 1:7.5 solid-liquid ratio, the initial PH 6.5, inoculums amount 2% and culture temperature 28C, cellulase activity after 96 hrs culture was 2.11 u/ml, 3.5 folds higher than that of original strain.
