The different expression of KIT Gene in Liaoning new breed of cashmere goats with different color
KIT encodes growth factor receptors which can expess the precuror of the melanophore.It has also been known to play an important role in the multiplication, migration and survival of the melanophore.However, there are no research reports the diverse expression of KIT gene and its coding protein in goats with different color.The presence of KITmutation and KIT protein expression was examined in the white cashmere and black cashmere goats.A single exon 13 missence nucleotide A→ G was identified in cashmere goats with different color.Only histidine (H) →arginine (R) amino acid (AA) change was detected at KIT-exon 13 in white cashmere goat and black cashmere goat.Moreover, comparison with other species showed: there were mainly three dramatic amino acid mutations areas.Our results also showed that the expression of the c-kit in the white Cashmere goat was higher than the black one which has significantly difference by the result of q-PCR as well as western blot.All cashmere goats with different color available for immunhistochemical analysis showed either weak (the black Cashmere goat) or strong (the white Cashmere goat) expression of the KIT protein, these findings suggested that there had a relationship between the mutation of KIT-exon 13 and the cashmere goats with different color.That could lay the foundation for the further research of the KIT-exonl 3 and color controllability regulation of the cashmere goats.
