Dietary Diacylglycerol Suppresses Appetite by Affecting Gene Expression of NPY, AgRP and POMC in Rats

  The inhibitory effects of dietary diacylglycerol (DG) on lipid accumulation,appetite and the expression of several hypothalamic genes related to energy homeostasiswere were investigated in this paper.Male Sprague-Dawley rats were randomly selected and given free access to diets of 5% (wt) triacylglycerol (TG,control group),20% (wt) TG or 20% (wt) DG diets for 10 weeks.At the end of experiment,the rats were sacrificed,the ratio of lipids to body weight were calculated,and the blood glucose,serum lipids,blood insulin,blood leptin were measured.Reverse transcriptase–polymerase chain reaction (RT-PCR) was used for measuring the expression of neuropeptide Y (NPY),agouti-related peptide (AgRP) and precursor pro-opiomelanocortin (POMC) mRNA in the hypothalamus.DG diets results in significant restraint of body weight gain,visceral fat,ratio of abdominal fat to body weight,increase and serum triglyderide,glucose,insulin and leptin levels comparing to High TG diets; Dietary diacylglycerol suppressed the energy intake increasing in rats compared with TG group.These were accompanied by the down-regulation of NPY and AgRP mRNA levels,and up-regulation of POMC mRNA levels.These results suggest that diacylglycerol improved fat metabolism and reduced fats accumulation may be associated with improved appetite control and energy balance.
