HPLC fingerprint combined with Self-reference method and Between-group method for pattern recognition of the dissolution of Tall gastrodia capsule
Gastrodiae Rhizoma (Chinese name: Tianma) is the dried rhizomes of Gastrodia elata Blume (Orchidaceae) and extremely valuable Traditional Chinese Medicine (TCM).Nowadays,with the development of modern pharmaceutical technology,it were crushed,sterilized and packaged into capsule as the Tall gastrodia capsule.Due to its precious herbal and complicated multi-component the evaluation method for TCM preparation is very significant.To evaluate the quality of Tall gastrodia capsule,the dissolution of it was evaluated based on chromatographic fingerprint combined with different method of pattern recognition.Self-reference method (defined as method A) could judge whether the main constituents were simultaneously released or not,calculate both the gross release and the respective dissolution rates of each batch.While method A can not compare with the quality of different batches(eg.compared with the quality of batch 1 and batch 2).So another method of pattern recognition was used.Between-group method (defined as method B) could not only classify and distinguish the 10 batches,but also build a predictive model for each batch for discriminating if they were correctly classified.With method A,the main constituents were simultaneously not released,additionally 3 important indicators for dissolution quality control were confirmed as gastrodin,parishin and 4-hydroxybenzyl alcohol.Mehod B results demonstrated that ten batches were discriminated in 3 groups from their own manufacturers.Meanwhile,according to discriminant functions 1-10,98.3% of original grouped cases were correctly classified.Combined with two methods,it is possible to provide a powerful tool and the extensive prospect for the building and evaluating the quality of TCM.
